چکیده

The genome profile of exon-intron distributions is presented for six eukaryotic genomes (H. sapiens, P. troglodytes, M. musculus, D. rerio, C. elegans, D. melanogaster) to deduce similarities and differences among their genome design and architecture. Interestingly, in all the six genomes, the total length in exons, introns and intergenic DNA on each chromosome is significantly correlated to the chromosome size. Another correlation is found between the total intron length and total exon length for each genome. The results presented provide a framework for understanding the nature and patterns of exon-intron length distributions, the constraints on them and their role in genome design and evolution and provide an insight into the evolution of intron-exon structures of eukaryotic genes.

Birth of New Spliceosomal Introns in Fungi by Multiplication of Introner-like Elements

Spliceosomal introns are noncoding sequences that separate exons in eukaryotic genes and are removed from pre-messenger RNAs by the splicing machinery. Their origin has remained a mystery in biology since their discovery because intron gains seem to be infrequent in many eukaryotic lineages. Although a few recent intron gains have been reported, none of the proposed gain mechanisms can convinci...

Microsatellites in different eukaryotic genomes: survey and analysis.

We examined the abundance of microsatellites with repeated unit lengths of 1-6 base pairs in several eukaryotic taxonomic groups: primates, rodents, other mammals, nonmammalian vertebrates, arthropods, Caenorhabditis elegans, plants, yeast, and other fungi. Distribution of simple sequence repeats was compared between exons, introns, and intergenic regions. Tri- and hexanucleotide repeats prevai...

Comparative Analysis of the Exon-Intron Structure in Eukaryotic Genomes

The exon numbers and lengths vary in different eukaryotic species. With increasing completed genomic sequences, it is indispensable to reanalyze the gene organization in diverse eukaryotic genomes. We performed a large-scale comparative analysis of the exon-intron structure in 72 eukaryotic organisms, including plants, fungi and animals. We confirmed that the exon-intron structure varies massiv...

The architecture of pre-mRNAs affects mechanisms of splice-site pairing.

The exon/intron architecture of genes determines whether components of the spliceosome recognize splice sites across the intron or across the exon. Using in vitro splicing assays, we demonstrate that splice-site recognition across introns ceases when intron size is between 200 and 250 nucleotides. Beyond this threshold, splice sites are recognized across the exon. Splice-site recognition across...

First Exons and Introns - A Survey of GC Content and Gene Structure in the Human Genome

Most transcriptional regulatory elements are located in non-coding DNA. In particular, some first introns play a vital role in transcriptional control and splicing. The length and GC-content of first exons and introns in complex organisms suggests that these structural units are likely to be important functional elements in large genomes. Hence, in this paper we perform a systematic comparison ...
